Checking transformation efficiency for different Lagenaria siceraria genotypes by using seed germination pouches as a growth carrier

Abstract Bottle gourd [ Lagenaria siceraria (Mol.) Standl.] is one of the most common horticultural and medicinal crops. Genetic transformation a rate-limiting step in unraveling function genes molecular breeding this non-model crop. In study, we established an optimized protocol for using seed germination pouch as growth carrier developed bottle composite plants. Overground explants were inoculated with Agrobacterium rhizogenes strain R1000, which harbored binary vector pBI121 carrying GUS reporter gene. The integration into transgenic roots was confirmed via reverse transcription-polymerase chain reaction histochemical staining. Furthermore, screened desirable genotypes from 14 germplasm collections based on number induced roots, efficiency, induction rate, “Changguang” (transformation approximately 36.8%) selected genotype suitable hairy root gourd. presented here rapid, suitable, efficient tool studying biology future.
